In vitro assays for evaluating the cellular responses to DNA damage induced by solar UV

چکیده

Solar UV induces dimeric photolesions in the genome of human skin cells, which potentially cause cutaneous abnormalities including cancer and aging. Normal human cells possess various systems to protect their genome from the harmful DNA lesions and nucleotide excision repair plays a dominant role in removing and fixing those DNA lesions. Measuring DNA lesions induced in the cells as well as monitoring the activity of cellular repair activity are helpful for evaluating the risk suffered from solar UV. We have previously established several monoclonal antibodies specifically recognizing UV-induced DNA lesions and developed in vitro immunoassays using the antibodies. Here I show various applications of the immunoassays to evaluate the cellular responses to UV-induced DNA lesions.

Antioxidant capacity and radioprotective properties of the flavonoids galangin and kaempferide isolated from Alpinia galanga L. (Zingiberaceae) against radiation induced cellular DNA damage

Background: Alpinia galanga L. belonging to the family Zingiberaceae is widely grown in the state of Kerala, India. They are effective antioxidant and free radical scavenger under both in vitro and in vivo condition. The efficacy of the isolated flavonoids in conferring protection from radiation induced damages to genomic DNA was studied. Materials and Methods: The bioflavonoids, galangin and k...

DNA Damage in Leukocytes from Fanconi Anemia (FA) Patients and Heterozygotes Induced by Mitomycin C and Ionizing Radiation as Assessed by the Comet and Comet-FISH Assay

Background: Lymphocytes of Fanconi anemia (FA) show an increased sensitivity to the alkylating agents such as mitomycin C (MMC), but their responses to gamma-irradiation is controversial. The extent of DNA damage in leukocytes of FA patients following irradiation and MMC treatment was studied at cellular and single chromosome level. Methods: DNA damage induced by gamma-rays and MMC was measure...

Characterization of Immune Responses Induced by Combined Clade-A HIV-1 Recombinant Adenovectors in Mice

Background: Numerous evidences indicate that in some HIV-1 positive patients, the humoral and cellular immune responses are induced against HIV-1 proteins and this is inversely related to the progress of infection. Objective: The aim of this study was the evaluation of the Adenovectors containing HIV genes in induction of immune responses in mice. Methods: The HIV-1 genes including gag p24, rev...
